Description

Monoclonal antibody C10 recognizes human lysosome-associated membrane protein-2 (LAMP-2; CD107b). LAMP-2 a heavily glycosylated membrane protein expressed in lysosomes. It is a member of a family of membrane glycoproteins like MAP-1, LIMP2 and DC-LAMP. It is a heavily glycosylated protein of 383 amino acids. LAMP-2 is expressed in a broad panel of tissues like placenta, lung, liver and kidney. Glycosylation is dependent on cell type and activation status. LAMP-2 exists in three splice variants LAMP-2A, LAMP-2B and LAMP-2C. They differ in their sequence in the transmembrane and cytoplasmic domain that effects their function. LAMP-2 is important for control of lysosomal activity and traffic across the lysosomal membranes. Its ability to mediate fusion between autophagosomes and lysosomes makes it important for the process of autophagy. The protein is also involved in cholesterol transport and cytoplasmic antigen MHCII presentation. Normally LAMP-2 is expressed at low levels on the cell surface. The degree of surface expression is dependent on the cell type and state of activation. Genetic deficiency of LAMP-2 causes Danon disease inducing cardiomyopathy and myopathy.
